Current evaluation Conclusion from the previous Monograph: Tetrachloroethylene (Perc) is probably carcinogenic to humans (Group 2A) with the working group finding limited evidence in humans for the carcinogenicity of Perc and sufficient evidence in experimental animals for the carcinogenicity of Perc. In making the overall evaluation, the Working Group considered the following evidence: (i) Although Perc is known to induce peroxisome proliferation in mouse liver, a poor quantitative correlation was seen between peroxisome proliferation and tumor formation in the liver after administration of Perc by inhalation. The spectrum of mutations in proto-oncogenes in liver tumors from mice treated with Perc was different from that in liver tumors from mice treated with trichloroethylene (TCE). (ii) The compound induced leukemia in rats. (iii) Several epidemiological studies showed elevated risks for esophageal and cervical cancer and nonHodgkin lymphoma, which the working group stated was unlikely due to chance although confounding could not be excluded.
